Partition Chromatography of Organic Acids in Body Fluids with Silica Gel Applicafion of the Method to Normal Human Urine Samuel Meites IN 1941, MnTIN AND SYNGE, in their now classical paper, introduced silica gel as an inert stationary phase in the partition chromatog- raphy of acetylated amino acids (1). The use of silica gel was sub- sequently extended to the separation of straight chain saturated fatty acids (2-5). Isherwood (6) introduced silica gel as a medium for the separation of various volatile and non-volatile acids, particu- larly in fruit, as well as successfully using it for the extraction of the acids. Isherwood’s method was subsequently extended and modified by several workers (7-13). Frohman, Orten, and Smith (9) separated acids of the citric acid cycle from rat tissues with the aid of silica gel columns. They were able to report quantitative values for lactic, citric and pyruvic acids in blood. In this paper, use of the silica gel column for the separation of non-a-amino organic acids in human urine is described. METHOD OF EXTRACTION The method of Isherwood may be successfully applied to the extrac- tion of the organic acids in urine. Preliminary unpublished results indicate that the extraction procedure is also applicable to sweat, saliva, spinal, and amniotic fluids. For urine the method routinely followed has been to collect a 24-hour specimen, the urine being stored From the Clinical Laboratory, Children’s Hospital, and the Department of Pediatrics, Ohio State Univer8ity College of Medicine, Columbus, Ohio.
